Filmin konusu 19. yy'ın sonlarında geçmektedir. Amcası Sir Charles Baskerville'in esrarengiz bir hayvan tarafından parçalanarak öldürülmesi üzerine İngiltere'nin Devonshire kırsalındaki tarihi malikâneyi devralmak üzere buraya gelen yeğen Sir Henry Baskerville (Christopher Lee), 1700'lü yıllarda ataları olan kötü namlı Sir Hugo Baskerville (David Oxley)'in de aynı şekilde ölmüş olduğunu hatırlar. Ailenin üzerinde bir lânet olduğuna inanılmaktadır. Hayatının tehlikede olduğunu düşünerek Londra'dan ünlü dedektif Sherlock Holmes (Peter Cushing) ve ortağı Doktor Watson (André Morell)'u yardıma çağırır. Zeki dedektif her zaman olduğu gibi kendine has yöntemlerle olayı aydınlatacaktır. Romandan ufak tefek bazı farklılıkları olmasına rağmen filmin kaynağına oldukça sadık bir sinema uyarlaması olduğu belirtilmiştir..
